Genetic Improvement to Obtain Specialized Haematococcus pluvialis Genotypes for the Production of Carotenoids, with Particular Reference to Astaxanthin
Nowadays, the search for natural substances with a high nutraceutical effect positively
impact the world market. Among the most attractive macromolecules are antioxidants, capable
of preventing the development of various pathologies. Astaxanthin (ASX) is antioxidant molecule
produced by the microalga H. pluvialis as a response to different types of stress. Usually, astaxanthin
production involves the first phase of accumulation of the biomass of H. pluvialis (green phase), which
is then stressed to stimulate the biosynthesis and accumulation of ASX (red phase). In this study,
the H. pluvialis wild-type strain was subjected to random mutagenesis by UV. Among the different
mutant strains obtained, only two showed interesting bio-functional characteristics, such as a good
growth rate. The results demonstrated that the HM1010 mutant not only has a higher growth trend
than the WT mutant but accumulates and produces ASX even in the green phase. This innovative
genotype would guarantee the continuous production of ASX, not linked to the two-step process and
the uniqueness of the product obtained
Modeling Events and Interactions through Temporal Processes -- A Survey
In real-world scenario, many phenomena produce a collection of events that
occur in continuous time. Point Processes provide a natural mathematical
framework for modeling these sequences of events. In this survey, we
investigate probabilistic models for modeling event sequences through temporal
processes. We revise the notion of event modeling and provide the mathematical
foundations that characterize the literature on the topic. We define an
ontology to categorize the existing approaches in terms of three families:
simple, marked, and spatio-temporal point processes. For each family, we
systematically review the existing approaches based based on deep learning.
Finally, we analyze the scenarios where the proposed techniques can be used for
addressing prediction and modeling aspects.Comment: Image replacement
Delucidazione del profilo di glucosinolati in Eruca sativa L. mediante Spettrometria di B18 Massa Ciclotronica in Trasformata di Fourier (FTICR-MS)
I glucosinolati (GLSs) sono metaboliti secondari presenti in tutte le piante dell’ordine Capparales, che tra le altre famiglie include le Brassicaceae alla quale appartengono note specie come le rape, i broccoli e la rucola. Oltre ad essere utilizzati dalle piante come deterrenti contro gli insetti nocivi e quindi come agrofarmaci naturali, questi composti, ed in particolare i loro prodotti di degradazione, presentano una serie di interessanti proprietà biologiche. Alcuni di essi hanno effetti negativi sulla salute umana, influenzando il funzionamento della ghiandola tiroidea, altri invece hanno effetti benefici. Evidenze epidemiologiche suggeriscono che l’assunzione dei vegetali della Brassica mediante dieta è associata ad un ridotto rischio di cancro. Poiché le specifiche proprietà biologiche dei glucosinolati variano considerevolmente in funzione della loro tipologia, risulta di particolare importanza identificare e quantificare accuratamente tali composti nei tessuti vegetali. La complessità strutturale dei diversi glucosinolati rende necessaria la disponibilità di metodi analitici grazie ai quali sia possibile una loro caratterizzazione strutturale.
La cromatografia liquida con rivelazione di massa (LC-MS) si sta mostrando una tecnica particolarmente efficace per l’identificazione di composti non noti estratti da prodotti naturali. L’accoppiamento LC-MS permette, infatti, di sfruttare simultaneamente la grande efficienza separativa propria della HPLC e la rilevante capacità di identificazione della MS. Il lavoro svolto dal nostro gruppo di ricerca negli ultimi anni ha confermato le potenzialità della cromatografia liquida accoppiata alla spettrometria di massa a trappola ionica con ionizzazione elettrospray (LC-ESI-ITMS) nella determinazione rapida ed efficace del profilo di GAs e GLSs negli estratti di piante. Tuttavia, i ragguardevoli progressi tecnologici che negli ultimi anni hanno permesso lo sviluppo di tecniche ad alta risoluzione come la spettrometria di massa ciclotronica in trasformata di Fourier (FTICR-MS), hanno facilitato notevolmente la delucidazione delle strutture di molti metaboliti secondari delle piante, anche se presenti in quantità molto basse. Poiché non tutti i GLSs sono stati ad oggi identificati, in questo lavoro abbiamo voluto verificare le potenzialità della FTICR-MS nella delucidazione del profilo di questi metaboliti secondari in estratti grezzi di foglie di rucola.
L’alta risoluzione e l’elevata accuratezza di massa che contraddistinguono questa tecnica hanno portato a determinare la presenza di due nuovi glucosinolati che, seppur noti, mai erano stati identificati in foglie di rucola
